Resulta que el virus del herpes simple-1 (VHS-1) tambi\u00e9n redecora el interior de las c\u00e9lulas, seg\u00fan un nuevo estudio publicado en Nature Communications.<\/p>\n<p>Un equipo del Centro de Regulaci\u00f3n Gen\u00f3mica (CRG) de Barcelona ha descubierto que el virus del herpes labial remodela la arquitectura del genoma humano, reorganizando su forma en el espacio tridimensional para que el VHS-1 pueda acceder a los genes del hu\u00e9sped m\u00e1s \u00fatiles por su capacidad de reproducci\u00f3n.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.agenciasinc.es\/bundles\/app\/images\/quotebefore.png\" \/><\/p>\n<p>VHS-1 es un interiorista oportunista que remodela con precisi\u00f3n el genoma humano para aprovechar mejor los recursos del hu\u00e9sped, un mecanismo de manipulaci\u00f3n que no conoc\u00edamos<\/p>\n<p>Esther Gonz\u00e1lez Almela, primera firmante (CRG)<br \/>\n<img decoding=\"async\" src=\"https:\/\/www.agenciasinc.es\/bundles\/app\/images\/quoteafter.png\" \/><\/p>\n<p>\u00abVHS-1 es un interiorista oportunista que remodela el genoma humano con gran precisi\u00f3n y elige con qu\u00e9 partes entra en contacto. Es un nuevo mecanismo de manipulaci\u00f3n que no sab\u00edamos que ten\u00eda el virus para aprovechar los recursos del hu\u00e9sped\u00bb, afirma Esther Gonz\u00e1lez Almela, primera firmante del estudio.<\/p>\n<p>Si bien se ha visto que otros virus del herpes compactan y remodelan los cromosomas del hu\u00e9sped, no estaba claro si era un efecto secundario de la invasi\u00f3n del virus y la creaci\u00f3n de sus propias f\u00e1bricas de replicaci\u00f3n viral.<\/p>\n<p><a><\/a><\/p>\n<h2>Remodelaci\u00f3n deliberada del genoma<\/h2>\n<p>Seg\u00fan los autores, este estudio es la primera prueba que demuestra que el VHS-1 remodela el genoma humano deliberadamente y a las pocas horas de la infecci\u00f3n.<\/p>\n<p>Un hallazgo importante del estudio es que el equipo demostr\u00f3 que la inactivaci\u00f3n de una sola enzima humana, la topoisomerasa I, bloquea por completo la capacidad del HSV-1 para reorganizar el genoma humano durante la infecci\u00f3n, lo que detiene la toma de poder hostil. El descubrimiento representa una nueva estrategia para controlar un virus que infecta a casi cuatro mil millones de personas en todo el mundo.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.agenciasinc.es\/bundles\/app\/images\/quotebefore.png\" \/><\/p>\n<p>En cultivo celular, la inhibici\u00f3n de la enzima, la topoisomerasa I,\u00a0detuvo la infecci\u00f3n antes de que el virus pudiera producir una sola part\u00edcula nueva.\u00a0Esto proporciona una diana terap\u00e9utica para detener la infecci\u00f3n<\/p>\n<p>Pia Cosma, autora principal (CRG)<br \/>\n<img decoding=\"async\" src=\"https:\/\/www.agenciasinc.es\/bundles\/app\/images\/quoteafter.png\" \/><\/p>\n<p>\u00abEn cultivo celular, la inhibici\u00f3n de esta enzima detuvo la infecci\u00f3n antes de que el virus pudiera producir una sola part\u00edcula nueva\u00bb, explica la profesora de investigaci\u00f3n ICREA Pia Cosma, autora principal del estudio en el CRG. \u00abEsto proporciona una nueva y potencial diana terap\u00e9utica para detener la infecci\u00f3n\u00bb.<\/p>\n<p>Los hallazgos fueron posibles combinando microscop\u00eda de superresoluci\u00f3n, una t\u00e9cnica de imagen que puede ver estructuras de 20 nan\u00f3metros de ancho (algo aproximadamente 3.500 veces m\u00e1s fino que un cabello), con Hi-C, una t\u00e9cnica que revela qu\u00e9 fragmentos de ADN se est\u00e1n tocando dentro del n\u00facleo. Utilizaron ambas t\u00e9cnicas para entender los mecanismos que usa VHS-1 para secuestrar a las c\u00e9lulas humanas.<\/p>\n<p><a><\/a><\/p>\n<h2>Sofisticada estrategia de ataque<\/h2>\n<p>Descubrieron que la toma de poder hostil comienza durante la primera hora de invasi\u00f3n viral. El virus secuestra a la enzima ARN-polimerasa II humana para que le ayude a sintetizar sus propias prote\u00ednas. La topoisomerasa I, una enzima que corta el ADN para liberar el estr\u00e9s torsional, y la cohesina, una prote\u00edna estructural, siguieron a la ARN-polimerasa II humana a los compartimentos de replicaci\u00f3n viral reci\u00e9n formados.<\/p>\n<p>La toma de poder hostil comienza durante la primera hora de invasi\u00f3n viral. El virus secuestra a la enzima ARN-polimerasa II humana para que le ayude a sintetizar sus propias prote\u00ednas.<\/p>\n<p>Tres horas despu\u00e9s de la infecci\u00f3n, la mayor\u00eda de las polimerasas y una fracci\u00f3n considerable de los otros dos factores hab\u00edan abandonado los genes humanos.<\/p>\n<p>El hurto molecular hace que la transcripci\u00f3n se venga abajo en todo el genoma del hu\u00e9sped, lo que a su vez provoca que la cromatina, el estado natural del genoma humano dentro de las c\u00e9lulas, se compacte en una c\u00e1scara densa de solo el 30 % de su volumen original.<\/p>\n<p><a><\/a><\/p>\n<h2>Hallazgo inesperado<\/h2>\n<p>Este fue un hallazgo inesperado, ya que se cre\u00eda que la estructura de la cromatina dicta la transcripci\u00f3n. \u00abSiempre pensamos que la cromatina densa apagaba los genes, pero aqu\u00ed vemos lo contrario: primero se detiene bastante la transcripci\u00f3n y luego el ADN se compacta. La relaci\u00f3n entre actividad y estructura podr\u00eda ser una calle de doble sentido\u00bb, dice \u00c1lvaro Castells Garc\u00eda, primer coautor del estudio e investigador del CRG.<\/p>\n<p>El virus del herpes labial infecta de por vida a dos de cada tres personas menores de 50 a\u00f1os. No tiene cura y las cepas resistentes a los tratamientos son cada vez m\u00e1s frecuentes<\/p>\n<p>Dos de cada tres personas menores de 50 a\u00f1os viven con el VHS-1. Una vez infectadas, las personas tienen el virus de por vida, aunque la mayor\u00eda de los casos son asintom\u00e1ticos o se manifiestan como herpes labial recurrente. En raras ocasiones, el virus puede causar ceguera o enfermedad potencialmente mortal en reci\u00e9n nacidos y personas inmunodeprimidas.<\/p>\n<p>Los hallazgos del estudio pueden ayudar a abordar la carga que el VHS-1 representa para la salud p\u00fablica, ya que se considera un desaf\u00edo para la salud mundial debido a su prevalencia y capacidad para causar brotes recurrentes.
