En el proyecto tambi\u00e9n ha participado el Servicio de Oncolog\u00eda y de Patolog\u00eda del Hospital del Mar, junto con investigadores del Grupo GEICAM de Investigaci\u00f3n en C\u00e1ncer de Mama.<\/p>\n<p>TIM-3 permite a las c\u00e9lulas tumorales\u00a0suprimir el sistema inmunitario\u00a0cuando llegan a los \u00f3rganos distantes y as\u00ed asegurar su supervivencia<\/p>\n<p>El trabajo, que publica Cancer Cell, determina el papel de la prote\u00edna TIM-3 en las c\u00e9lulas m\u00e1s agresivas iniciadoras de met\u00e1stasis, las llamadas semillas de la met\u00e1stasis. En ellas, TIM-3 permite a las c\u00e9lulas tumorales suprimir el sistema inmunitario cuando llegan a los \u00f3rganos distantes y as\u00ed asegurar su supervivencia en un momento cr\u00edtico. Este mismo hecho convierte a TIM-3 en una diana terap\u00e9utica \u00fanica y espec\u00edfica para este preciso momento de la enfermedad.<\/p>\n<p>Esta etapa, la de la llegada al nuevo \u00f3rgano, es cr\u00edtica, pero poco conocida porque no hay manifestaciones cl\u00ednicas ni t\u00e9cnicas que permitan investigarlo. El equipo responsable del trabajo ha utilizado un modelo de rat\u00f3n para hacerlo y describir el papel clave de TIM-3 en la supervivencia de las primeras c\u00e9lulas tumorales que siembran los \u00f3rganos afectados.<\/p>\n<p>\u201cSe trata de una fase de vulnerabilidad de la enfermedad, ya que quedan pocas c\u00e9lulas en el lugar. Si entendemos qu\u00e9 pasa y somos capaces de erradicarlas, dispondremos de una gran oportunidad para evitar la met\u00e1stasis cl\u00ednica, que es la fase peligrosa de la enfermedad\u201d, explica el Toni Celi\u00e0-Terrassa, firmante principal del estudio e investigador del Grupo GEICAM.<\/p>\n<p><a><\/a><\/p>\n<h2>Evitar el ataque inmunitario<\/h2>\n<p>Las c\u00e9lulas cancerosas que sobreviven al viaje desde el tumor primario utilizan TIM-3 para reconfigurar el sistema inmunitario del nuevo \u00f3rgano y evitar su ataque. Esto les permite proliferar y generar un nuevo tumor, la iniciaci\u00f3n de la met\u00e1stasis, como han podido ver los investigadores con c\u00e9lulas de los diversos tipos de c\u00e1ncer de mama en met\u00e1stasis en el h\u00edgado.<\/p>\n<p>Las pacientes con tumores con niveles altos de TIM-3 ten\u00edan m\u00e1s riesgo de met\u00e1stasis y de mal pron\u00f3stico<\/p>\n<p>Adem\u00e1s, con una cohorte de pacientes del Hospital del Mar pudieron comprobar como aquellas pacientes con tumores con niveles altos de TIM-3 ten\u00edan m\u00e1s riesgo de met\u00e1stasis y de mal pron\u00f3stico. \u201cLos pacientes positivos en TIM-3 en el tumor primario tienen un riesgo m\u00e1s elevado de recurrencia y met\u00e1stasis\u201d, apunta Catalina Rozal\u00e9n, investigadora predoctoral del Instituto de Investigaci\u00f3n del Hospital del Mar.<\/p>\n<p>Esto propone la posibilidad de utilizar f\u00e1rmacos existentes que bloqueen la acci\u00f3n de TIM-3 para evitar la aparici\u00f3n de nuevos tumores. \u201cTener herramientas para estos pacientes de alto riesgo, como los que son TIM-3 positivo, puede permitir aplicarles una terapia anti-TIM-3 despu\u00e9s de la cirug\u00eda para evitar la posterior reca\u00edda y prevenir la met\u00e1stasis\u201d, dice Celi\u00e0-Terrassa.<\/p>\n<p>Este estudio precl\u00ednico indica que habr\u00eda que aplicar el tratamiento antes de la aparici\u00f3n de la met\u00e1stasis, ya que cuando ya se ha producido la enfermedad es m\u00e1s dif\u00edcil de controlar y TIM-3 deja de tener la misma relevancia.
