En lugar de dirigirse a las neuronas, el tratamiento act\u00faa sobre la barrera hematoencef\u00e1lica (BBB), la interfaz que regula el entorno del cerebro.<\/p>\n<p>En lugar de emplear nanopart\u00edculas como veh\u00edculos de f\u00e1rmacos, este m\u00e9todo utiliza part\u00edculas bioactivas por s\u00ed mismas<\/p>\n<p>Al reparar esta barrera, que funciona como guardi\u00e1n frente a toxinas y pat\u00f3genos, los cient\u00edficos lograron restaurar el equilibrio del sistema vascular cerebral y revertir la patolog\u00eda en modelos animales.<\/p>\n<p>El cerebro consume un 20 % de la energ\u00eda en adultos y hasta un 60 % en ni\u00f1os, a trav\u00e9s de un denso entramado de capilares. Cuando la barrera hematoencef\u00e1lica se ve comprometida, el sistema de limpieza cerebral deja de funcionar correctamente, lo que favorece la acumulaci\u00f3n de la prote\u00edna beta amiloide (A\u03b2), una de las principales caracter\u00edsticas del alzh\u00e9imer.<\/p>\n<p><a><\/a><\/p>\n<h2>Directas al torrente sangu\u00edneo<\/h2>\n<p>Los investigadores lograron que las nanopart\u00edculas permitieran el paso de estas prote\u00ednas de desecho al torrente sangu\u00edneo para su eliminaci\u00f3n. En modelos de rat\u00f3n programados gen\u00e9ticamente para producir grandes cantidades de A\u03b2 y desarrollar deterioro cognitivo, la administraci\u00f3n de solo tres dosis consigui\u00f3 resultados notables.<\/p>\n<p>En modelos de rat\u00f3n modificados para producir grandes cantidades de A\u03b2 y desarrollar deterioro cognitivo, la administraci\u00f3n de solo tres dosis consigui\u00f3 resultados notables<\/p>\n<p>\u201cSolo una hora despu\u00e9s de la inyecci\u00f3n observamos una reducci\u00f3n del 50-60 % en la cantidad de A\u03b2 dentro del cerebro\u201d, explica Junyang Chen, primer coautor del estudio e investigador del Hospital West China y del University College London.<\/p>\n<p>M\u00e1s all\u00e1 de la eliminaci\u00f3n inmediata de prote\u00ednas t\u00f3xicas, el equipo observ\u00f3 efectos terap\u00e9uticos\u00a0duraderos. En uno de los experimentos, un rat\u00f3n de 12 meses (equivalente a un humano de 60 a\u00f1os) recibi\u00f3 el tratamiento y fue analizado seis meses despu\u00e9s. A los 18 meses, edad comparable a los 90 a\u00f1os en humanos, el animal mostr\u00f3 un comportamiento similar al de un rat\u00f3n sano.<\/p>\n<p>Un rat\u00f3n tratado a los 12 meses recuper\u00f3, medio a\u00f1o despu\u00e9s, el comportamiento de un animal sano pese a tener una edad equivalente a 90 a\u00f1os en humanos<\/p>\n<p>\u201cEl efecto a largo plazo proviene de la restauraci\u00f3n del sistema vascular del cerebro. Creemos que funciona como una cascada: cuando la vascularizaci\u00f3n recupera su funci\u00f3n, empieza a eliminar A\u03b2 y otras mol\u00e9culas da\u00f1inas, lo que permite que el sistema vuelva a equilibrarse. Nuestras nanopart\u00edculas act\u00faan como un f\u00e1rmaco y parecen activar un mecanismo de retroalimentaci\u00f3n que normaliza esta v\u00eda de eliminaci\u00f3n\u201d, afirma Giuseppe Battaglia, profesor de investigaci\u00f3n ICREA en el IBEC y l\u00edder del estudio.<\/p>\n<p><a><\/a><\/p>\n<h2>F\u00e1rmacos supramoleculares<\/h2>\n<p>En condiciones normales, la prote\u00edna LRP1 transporta la beta amiloide a trav\u00e9s de la barrera hematoencef\u00e1lica para su eliminaci\u00f3n. Este proceso puede bloquearse si la prote\u00edna se une en exceso, o ser insuficiente si la se\u00f1al es demasiado d\u00e9bil, en ambos casos facilitando la acumulaci\u00f3n en el cerebro.<\/p>\n<p>Los f\u00e1rmacos supramoleculares desarrollados imitan a los ligandos de LRP1, favoreciendo el transporte de A\u03b2 y restableciendo el sistema natural de limpieza.
