Un equipo del Centro de Investigaci\u00f3n del C\u00e1ncer (CSIC-USAL-FICUS) ha identificado el papel de una prote\u00edna en el desarrollo y la resistencia a los tratamientos de los tumores pulmonares que tienen mutaciones en los genes KRAS y TP53. Este tipo de c\u00e1ncer, cuyo D\u00eda Internacional se celebra cada 17 de noviembre, es uno de los m\u00e1s mortales.<\/p>\n

La prote\u00edna Capicua frena la transformaci\u00f3n maligna causada por estas alteraciones gen\u00e9ticas y act\u00faa como una barrera natural para la formaci\u00f3n de estos tumores<\/p>\n

El estudio,\u00a0que aparece publicado en la revista\u00a0EMBO Molecular Medicine<\/a>, demuestra que la prote\u00edna Capicua frena la transformaci\u00f3n maligna causada por estas alteraciones gen\u00e9ticas y act\u00faa, por tanto, como una barrera natural para la formaci\u00f3n de estos tumores.<\/p>\n

El laboratorio de\u00a0Matthias Drosten, especializado en mejorar la comprensi\u00f3n de la biolog\u00eda del c\u00e1ncer de pulm\u00f3n y el dise\u00f1o de tratamientos dirigidos a este tipo de c\u00e1ncer, ha comprobado que los tumores pulmonares pueden superar esa barrera natural a trav\u00e9s de ganancias en el n\u00famero de copias del gen KRAS. En este contexto, el equipo ha confirmado que la p\u00e9rdida de Capicua contribuye directamente a la resistencia a los tratamientos.<\/p>\n

La investigaci\u00f3n tambi\u00e9n abre nuevas v\u00edas terap\u00e9uticas: la restauraci\u00f3n de la actividad represora de Capicua o el ataque a factores que compensan su p\u00e9rdida pueden revertir la proliferaci\u00f3n tumoral y restaurar la sensibilidad a los medicamentos.<\/p>\n

\u201cHemos descubierto que Capicua es mucho m\u00e1s que un simple actor secundario en la v\u00eda oncog\u00e9nica de KRAS. Abre nuevas oportunidades para intervenir en fases tempranas de la enfermedad\u201d, subraya\u00a0Drosten. \u201cUna vez perdida su funci\u00f3n de represi\u00f3n, el crecimiento tumoral se dispara y, adem\u00e1s, aparecen resistencias a f\u00e1rmacos que antes eran efectivos\u201d, a\u00f1ade. \u200b<\/p>\n

Anticipar la resistencia<\/h2>\n

KRAS es uno de los oncogenes m\u00e1s estudiados y responsables del 30 % de los c\u00e1nceres humanos que, hasta hace poco, eran considerados \u201cintratables\u201d. Si bien existen terapias dirigidas actualmente aprobadas, la mayor\u00eda de los tumores acaban desarrollando resistencia.<\/p>\n

KRAS es uno de los oncogenes m\u00e1s estudiados y responsables del 30 % de los c\u00e1nceres humanos que, hasta hace poco, eran considerados \u201cintratables\u201d<\/p>\n

El equipo ha realizado experimentos con modelos animales, empleando ratones gen\u00e9ticamente modificados para replicar las mutaciones observadas en c\u00e1nceres humanos. Estas investigaciones permitieron estudiar de manera controlada el impacto de la p\u00e9rdida funcional de Capicua y de la amplificaci\u00f3n de KRAS en el desarrollo tumoral, as\u00ed como testar nuevas estrategias terap\u00e9uticas. Todos los protocolos con animales fueron aprobados por los comit\u00e9s \u00e9ticos pertinentes y cumplieron con las regulaciones internacionales sobre bienestar animal.<\/p>\n

Este descubrimiento tiene implicaciones importantes para la selecci\u00f3n y el tratamiento de los pacientes. La identificaci\u00f3n de mutaciones o alteraciones funcionales en Capicua permite anticipar la aparici\u00f3n de resistencias y dise\u00f1ar tratamientos personalizados basados en el perfil molecular de cada tumor.<\/p>\n

En este sentido,\u00a0Irene Ballesteros-Gonz\u00e1lez, primera autora del estudio, subraya: \u201cGracias a los modelos experimentales empleados en esta investigaci\u00f3n se han podido probar combinaciones farmacol\u00f3gicas. Adem\u00e1s, se ha demostrado que la reactivaci\u00f3n de Capicua, as\u00ed como el uso de inhibidores metab\u00f3licos espec\u00edficos, puede volver a sensibilizar los tumores resistentes a los f\u00e1rmacos convencionales\u201d. \u200b<\/p>\n

El tumor m\u00e1s mortal<\/h2>\n

Seg\u00fan los \u00faltimos datos del Registro de Tumores Tor\u00e1cicos del del Grupo Espa\u00f1ol de C\u00e1ncer de Pulm\u00f3n (GECP), este tipo de c\u00e1ncer sigue siendo el tumor m\u00e1s mortal, con 23 239 fallecimientos en 2024, un 1,9 % m\u00e1s que el a\u00f1o anterior. Adem\u00e1s, la mortalidad femenina crece un 7 %, y el c\u00e1ncer de pulm\u00f3n ya es la principal causa de muerte oncol\u00f3gica en mujeres.<\/p>\n

El tabaquismo sigue presente como causa directa de la enfermedad<\/p>\n

Los especialistas del GECP tambi\u00e9n alertan de nuevos perfiles: los diagn\u00f3sticos femeninos ya representan el 27,8 % y uno de cada veinte pacientes tiene menos de 50 a\u00f1os. El tabaquismo sigue presente como causa directa de la enfermedad: el 40,9 % es fumador y el 46 % exfumador.<\/p>\n

El c\u00e1ncer de pulm\u00f3n es el tumor m\u00e1s letal en Espa\u00f1a: s\u00f3lo 3 de cada 10 pacientes sobreviven a cinco a\u00f1os. Pese a ello, la supervivencia ha mejorado un 12 % en la \u00faltima d\u00e9cada gracias a la investigaci\u00f3n y las nuevas terapias. Sin embargo, s\u00f3lo el 4 % de la inversi\u00f3n p\u00fablica en c\u00e1ncer se destina al de pulm\u00f3n.<\/p>\n
