Cuando nuestro organismo combate una infecci\u00f3n, el sistema inmunitario debe activar defensas con rapidez y causar una inflamaci\u00f3n que es beneficiosa. Pero es igualmente importante resolver dicha inflamaci\u00f3n para volver a la normalidad. En ese equilibrio son esenciales los macr\u00f3fagos, c\u00e9lulas especializadas en fagocitar o engullir c\u00e9lulas que han muerto por la infecci\u00f3n viral y en reparar el da\u00f1o tisular asociado a la infecci\u00f3n o la reacci\u00f3n inflamatoria.<\/p>\n

Un estudio realizado en el Centro Nacional de Investigaciones Cardiovasculares Carlos III (CNIC) y publicado en la revista Immunity desvela el mecanismo por el que una se\u00f1al asociada a la respuesta inflamatoria y antiviral, el interfer\u00f3n de tipo I (IFN-I), ajusta las mitocondrias de los macr\u00f3fagos para impulsar la limpieza del da\u00f1o tisular y evitar una inflamaci\u00f3n descontrolada.<\/p>\n

El IFN-I, una citoquina que puede promover tanto respuestas inflamatorias como antiinflamatorias, regula la funci\u00f3n de los macr\u00f3fagos para resolver la inflamaci\u00f3n<\/p>\n

El IFN\u2011I es una citoquina capaz de impulsar respuestas inflamatorias o antiinflamatorias seg\u00fan el contexto de la enfermedad. Activa un programa espec\u00edfico de genes estimulados por interfer\u00f3n que permite a los macr\u00f3fagos eliminar con mayor eficacia las c\u00e9lulas muertas y reparar el da\u00f1o tisular asociado, por ejemplo, a infecciones v\u00edricas. El estudio reci\u00e9n publicado aborda c\u00f3mo el IFN\u2011I modula la funci\u00f3n de estas c\u00e9lulas para favorecer la resoluci\u00f3n de la inflamaci\u00f3n.<\/p>\n

El equipo del CNIC demuestra que, cuando los macr\u00f3fagos detectan \u00e1cidos nucleicos vinculados a infecciones v\u00edricas en su interior, ajustan su metabolismo. Sus mitocondrias reducen el potencial de membrana sin perder funcionalidad. Este cambio se desencadena por la producci\u00f3n de interfer\u00f3n tipo I (IFN\u2011I) en respuesta a la infecci\u00f3n, que se une a su receptor en la superficie del macr\u00f3fago y activa la s\u00edntesis de la prote\u00edna ISG15.<\/p>\n

Cambios metab\u00f3licos<\/h2>\n

\u201cHemos visto que una prote\u00edna espec\u00edfica estimulada por IFN\u2011I, llamada ISG15, se une a prote\u00ednas mitocondriales y provoca dos modificaciones coordinadas: un aumento en la producci\u00f3n de ATP y una reducci\u00f3n del potencial de membrana mitocondrial. Estos cambios metab\u00f3licos mejoran la capacidad de los macr\u00f3fagos para retirar c\u00e9lulas muertas, lo que facilita la resoluci\u00f3n de la inflamaci\u00f3n\u201d, explica la primera autora, Gillian Dunphy, investigadora del grupo de Inmunobiolog\u00eda dirigido por\u00a0David Sancho.<\/p>\n

Dunphy a\u00f1ade: \u201cAdem\u00e1s, la bajada del potencial de la membrana mitocondrial activa una proteasa que incrementa la fragmentaci\u00f3n de la mitocondria. Este proceso modifica el metabolismo y reduce la expresi\u00f3n de genes inflamatorios, de modo que el propio IFN\u2011I contribuye a poner fin a la inflamaci\u00f3n\u201d.<\/p>\n

La ca\u00edda del potencial mitocondrial activa una proteasa que fragmenta la mitocondria, ajusta el metabolismo del macr\u00f3fago y reduce la expresi\u00f3n de genes inflamatorios, de modo que el propio IFN\u2011I ayuda a frenar la inflamaci\u00f3n<\/p>\n

Para alcanzar estas conclusiones, los investigadores combinaron an\u00e1lisis metab\u00f3licos, microscop\u00eda avanzada y modelos experimentales celulares y animales. Observaron que el tratamiento con IFN\u2011I potencia la captaci\u00f3n de c\u00e9lulas apopt\u00f3ticas por los macr\u00f3fagos y que esta ventaja desaparece en ausencia de ISG15. Tambi\u00e9n comprobaron que los cambios en la forma y la din\u00e1mica de las mitocondrias act\u00faan como un regulador natural que evita un exceso de se\u00f1alizaci\u00f3n por interfer\u00f3n, fen\u00f3meno asociado a autoinflamaci\u00f3n en otros contextos.<\/p>\n

Equilibrar defensa y reparaci\u00f3n<\/h2>\n

David Sancho, director del laboratorio de Inmunobiolog\u00eda del CNIC, se\u00f1ala: \u201cEste hallazgo dibuja un nuevo mapa de c\u00f3mo las se\u00f1ales antivirales dialogan con el metabolismo celular para equilibrar defensa y reparaci\u00f3n. Comprender y modular este eje podr\u00eda inspirar estrategias para acelerar la resoluci\u00f3n de la inflamaci\u00f3n en diversas patolog\u00edas o ajustar tratamientos basados en interfer\u00f3n, maximizando beneficios y minimizando efectos indeseados\u201d.<\/p>\n
