Aunque en los \u00faltimos a\u00f1os han surgido f\u00e1rmacos capaces de bloquearlo, su efecto suele durar poco por la aparici\u00f3n de resistencias. Ahora, un estudio propone ir un paso m\u00e1s all\u00e1: hacer que la propia c\u00e9lula destruya esta prote\u00edna clave.<\/p>\n<p>El trabajo, liderado por el IRB Barcelona y el Centro de Investigaci\u00f3n del C\u00e1ncer (CSIC, USAL, FICUS), demuestra que destruir KRAS mutada, en lugar de bloquearla, induce una regresi\u00f3n m\u00e1s intensa del tumor en modelos de rat\u00f3n con adenocarcinoma de pulm\u00f3n. Publicado en Cancer Research, el estudio aporta adem\u00e1s una de las primeras descripciones en vivo de c\u00f3mo surgen resistencias frente a terapias basadas en degradaci\u00f3n dirigida.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.agenciasinc.es\/bundles\/app\/images\/quotebefore.png\" \/><\/p>\n<p>Los inhibidores de KRAS han supuesto un gran avance, pero las resistencias siguen siendo un problema. Ahora podemos ir un paso m\u00e1s all\u00e1 y hacer que la c\u00e9lula tumoral degrade esta prote\u00edna<\/p>\n<p>Cristina Mayor-Ruiz (IRB Barcelona)<br \/>\n<img decoding=\"async\" src=\"https:\/\/www.agenciasinc.es\/bundles\/app\/images\/quoteafter.png\" \/><\/p>\n<p>\u201cLos inhibidores de KRAS han supuesto un cambio enorme en el tratamiento de algunos c\u00e1nceres, pero las resistencias siguen siendo un gran problema. Ahora estamos entrando en una nueva etapa, en la que no solo podemos inhibir KRAS, sino tambi\u00e9n hacer que la propia c\u00e9lula tumoral lo degrade. El uso secuencial o incluso en combinaci\u00f3n de ambas estrategias farmacol\u00f3gicas podr\u00eda marcar la diferencia\u201d, explica\u00a0Cristina Mayor-Ruiz, investigadora del IRB Barcelona.<\/p>\n<p><a><\/a><\/p>\n<h2>Hacer que la c\u00e9lula elimine KRAS<\/h2>\n<p>as terapias de degradaci\u00f3n dirigida emplean mol\u00e9culas conocidas como PROTACs que inducen la destrucci\u00f3n de prote\u00ednas concretas. A diferencia de los inhibidores cl\u00e1sicos, no bloquean la actividad de la diana, sino que la eliminan.<\/p>\n<p>Como a\u00fan no existen PROTACs capaces de degradar directamente la mutaci\u00f3n KRASG12V, el equipo dise\u00f1\u00f3 un modelo experimental: introdujo en las c\u00e9lulas tumorales una etiqueta molecular en la prote\u00edna que permite su reconocimiento y utiliz\u00f3 PROTACs desarrollados en el IRB Barcelona, en colaboraci\u00f3n con el grupo del qu\u00edmico Antoni Riera, que se unen a esa etiqueta y desencadenan su degradaci\u00f3n.<\/p>\n<p>As\u00ed lograron eliminar KRASG12V en ratones y observar una marcada regresi\u00f3n tumoral. Adem\u00e1s, el modelo puede adaptarse para estudiar otros oncogenes y tipos de c\u00e1ncer.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.agenciasinc.es\/bundles\/app\/images\/quotebefore.png\" \/><\/p>\n<p>Las c\u00e9lulas tumorales son extremadamente dependientes de KRAS. Cuando eliminamos la prote\u00edna mutada, dejan de proliferar y entran en apoptosis, incluso sin la intervenci\u00f3n del sistema inmunitario<\/p>\n<p>David Santamar\u00eda (Centro de Investigaci\u00f3n del C\u00e1ncer)<br \/>\n<img decoding=\"async\" src=\"https:\/\/www.agenciasinc.es\/bundles\/app\/images\/quoteafter.png\" \/><\/p>\n<p>\u201cLas c\u00e9lulas tumorales son extremadamente dependientes de KRAS. Los mecanismos detectados difieren de los descritos con inhibidores de KRAS.<\/p>\n<p>En lugar de mutaciones adicionales en KRAS o reactivaci\u00f3n de rutas de se\u00f1alizaci\u00f3n, las c\u00e9lulas tumorales alteran la maquinaria encargada de degradar prote\u00ednas.
